According to the combined therapy, the tumor weight is reduced by about 50%, the tumor growth multiplication time is prolonged by 1.5 times, the neutrophil level of peripheral blood and important organs is not influenced, and the liver and kidney toxicity of VNP20009 and derivatives or gene modified strains thereof is remarkably reduced.

The virK gene deletion mutant strain and the complementary strain thereof are constructed, and the virulence difference of the virK gene deletion mutant strain and the complementary strain is compared in an animal infection model, so that the virulence of the virK deletion strain is obviously reduced, and the virulence phenotype of the complementary strain can be recovered, which indicates that the virK gene plays an important role in regulation and control of the virulence of klebsiella pneumoniae. It is found for the first time that PhoP can regulate and control virK gene expression, and the virulence phenotype of klebsiella pneumoniae is affected through the regulation and control way, so that a new virulence regulation and control mechanism is disclosed, and a theoretical basis is provided for subsequent targeted intervention.
